Does anyone know if Vbox does support virtualising a 5.25" floppy drive? I don’t see such an option…

If you mean, does it support using a physical 5.25'' floppy drive and virtualizing its connection to a virtual machine, then I'm not sure. If it's a drive connected with a standard IDE-like floppy connector, then it should do, seeing as I have a 3.5'' USB floppy drive which works in VirtualBox VMs, and reportedly Windows all the way up to 7 supports 5.25'' floppy drives.
If you mean virtual floppy disks of 5.25'' capacities (such as 320KB and 360KB) then yes, VirtualBox supports those. You can select them just like any other 1.44MB or 720KB floppy image file.
